Immunogenicity of islets of different clinically relevant sources in a preclinical humanized mouse model (A05)

Instant blood-mediated inflammatory reaction (IBMIR) is a key factor responsible for the rejection of up to 70% of the graft after intraportal islet transplantation in the context of type 1 diabetes treatment. In the final funding period we will take advantage of our IBMIR characterisation in humanised mouse models, to analyze IBMIR in response to porcine islets of different donor sources according to their predicted clinical relevance for the near (A) and far (B) future. To this end we will determine the immunogenicity of islets by using established biomarkers for inflammation after the transplantation of adult porcine islets (API, A), and later neonate porcine islets (NPI, B). We further aim at adding novel IBMIR biomarkers based on the use of novel technologies and mouse models, and we will determine the role for necrotic donor cell death in IBMIR - with the intension to prevent IBMIR in the future.
