This project aims to examine, whether an additional chemical or enzymatic treatment by glycosidases, peptidases, DNAses well as surfactant or chelating agent, can improve a mechanical plaque-biofilm removal by using air-polishing devices. The plaque biofilm will be cultured in vitro on rough titanium discs (diameter 5 mm). Supragingival plaque serve as inoculum for biofilm cultivation. The cultivation period will be seven days.First, the biofilm-destabilizing effect will be investigated. Therefore, specimen after treatment with chemicals or enzymes will be wobbled by a shaker to generate mechanical stress (shear stress). The shaking will replace loosely biofilm mass. After shaking, the specimen will be analyzed by fluorescence-microscope methods to identify possible reduction of biofilm mass.Further, the combination of chemical treatment and mechanical treatment with an air-polishing device will be investigated. Here, only substances will be used, which are evaluated to be suitable within the first test part.This project is an essential part of optimizing a possible new strategy for treating peri-implantitis.
