Project A06 aims to examine the role of CD95 in determining cellular fate. Our hypothesis posits that interactions between lipids and CD95 influence the lipid composition of the cell membrane, subsequently impacting the behavior of other membrane receptors. We postulate that the disruption of this cooperativity results in cellular apoptosis, while close cooperativity promotes cell survival. To investigate this hypothesis, we will employ bulk and single-cell lipidomics, proteomics, single-molecule microscopy techniques, and cell assays. These experiments will be conducted using brain tumor cells, both with and without mutations in CD95, designed to disrupt the lipid-CD95 interactions.
