Phosphoinositides (PIPs) regulate membrane dynamics and cell signaling. In project A08 we aim to dissect the role of PI 3-phosphate as regulatory switches in the control of lysosome function. We capitalize on our finding that distinct PI 3-phosphates differentially regulate lysosome function and nutrient signaling to (i) identify novel lysosomal PI 3-phosphate-binding effector proteins and define their roles in controlling (ii) lysosomal nutrient signaling and protein turnover as well as (iii) lysosome position and dynamics. These studies will provide new insights into the mechanisms underlying PIP-based functional switches at lysosomes.
