Elucidating the role of Lrrc34 in the regulation of ribosome biogenesis and Oct4 in mouse and human pluripotent stem cells

In the frame of the presented grant it will be the focus to analyze thefunction of the gene Lrrc34 in the nucleolus of pluripotent stem cells.Up till know there is not much known about the gene: it is exclusivelyexpressed in stem cells and male germ cells and its protein ispredominantly located in the nucleolus. It is shown, that regulation ofLrrc34 expression resulted in differentially growing of embryonic stemcells and an altered expression of pluripotency genes (e.g. Oct4).Since the nucleolus is the place of ribosome biogenesis,the role of Lrrc34 in ribosome biogenesis will beinvestigated. Furthermore the regulation of and interaction with Oct4 will be further analysed. For this, in vitro assays (mouse and human) as well as the generation and characterization of a knock-out mouse model will be used.
