Functional and molecular analysis of the epithelial barrier of porcine Peyer's patches follicle-associated epithelium
Final Report Abstract
The majority of immune cells are located in the gastrointestinal tract, demonstrating that the gut-associated lymphoid tissue (GALT) plays a pivotal role in the immune defense of the body. The GALT is composed of single isolated lymphoid follicles and aggregated follicles called Peyer´s patches (PP) The epithelium in this specific location is named follicleassociated epithelium (FAE) and contains specialized microfold cells (M cells), which have the function of presenting antigens to the underlying lymph follicles by transcytosis to induce an immune response or tolerance against the antigens. Therefore, the paracellular barrier in this region is tighter to ensure the route through the M cells to prevent an uncontrolled uptake of antigens. The capability of the immune system to balance between potentially harmful and helpful immune response is crucial to prevent an overactive immune system that could lead to tissue damage. This complex regulation is disturbed in inflammatory bowel diseases (IBDs) such as Crohn´s disease and ulcerative colitis. The pro inflammatory cytokine Tumor necrosis factor alpha (TNF) and its receptors TNF receptor 1 (TNFR-1) and TNF receptor 2 (TNFR-2) play a crucial role in the disease progression in IBDs. This project aimed to characterize barrier determinants in porcine PP, neighboring tissue and cell models, namely tight junction proteins, and the effects of endogenous and exogenous effectors. Our project provided detailed insights in the effects of TNF, as well as the effects of beneficially, mainly secondary plant compounds including cannabidiol, berberine, quercetin, cinnamic acid, and caffeic acid on barrier integrity and regulation. During the funding period, 12 peer review original publications and a series of conference abstracts and presentations, and two promotions of doctoral students emerged.
