Dynamics of Genome Duplication Machines (A01)

Biochemical and genetic studies have elucidated the basic mechanisms of DNA replication. We now want to investigate how the activities of different enzymes and auxiliary factors constituting the replication module are coordinated in vivo. We will measure the interactions and live-cell kinetics of different components of replication modules using a combination of novel fluorescence microscopy techniques. Biochemical and live cell data will be used to develop, test and refine kinetic models for the dynamic composition, function and genome-wide coordination of replication modules that assure precise and complete duplication of the entire genome.
