Blockade of the pro-inflammatory P2X7 receptor has beneficial effects in various rodent disease models but P2X7 antagonists have so far failed in clinical studies. Using in situ cross-linking and mass spectrometry we have identified a range of potential interactors. In the final funding period, we will continue to identify and validate extra- and intracellular P2X7 interactors and investigate to which extend murine P2X7 function and signaling is comparable to the human situation.
