In project P24 we will generate new chemical reagents with which to modulate and study native TRP channels. We will develop photoswitchable reagents for modulating TRP channels in their physiological contexts with high spatiotemporal precision, both using classical approaches (sterically demanding photoswitches on known pharmacophores) and developing a novel, general approach (biolocalisation photocontrol). We will also synthesise photoaffinity label derivatives of TRP modulators to characterise their binding sites, and provide ongoing synthetic support to the CRC. P24 therefore promotes the general discovery, development, characterisation and use of modulators for native TRP channels.
