Lab-in-a-tube: non-linear dielectric spectroscopy for multiparametric bioanalysis

There have been many efforts in developing robust bio-analytical systems to identify, quantify and analyze biological samples, to diagnose a health status or to evaluate certain drugs with higher accuracy. However, most of the state-of-the-art systems rely on the use of labels and/or sample enrichment strategies, requiring more complicated, time consuming and expensive devices and protocols. Here, we propose the development of a new class of label-free microtransducers for nonlinear dielectric spectroscopy, with the purpose to analyze the activation process of immunological cells during the progression/regression of multiple sclerosis disease. For that, rolled-up microtubular capacitor structures will be fabricated and integrated into fluidic microchannels for sample handling and enhanced detection throughput. Such rolled-structure with multiple windings will contain spacers to define multiple channels across the tubular capacitor to increase the contact area of the analyte and therefore its sensitivity. The proposed system will allow the detection of the inherent properties of biological samples such as conductivity, permittivity, and size upon external stimuli (e.g. drugs, electric/magnetic fields). Due to the interaction between the sample and the applied electric field at frequencies in the range between 100 MHz and 100 GHz, the discrimination between extra and intracellular molecular changes can be achieved with high selectivity and sensitivity. Finally, addressing the nonlinear and linear features of the obtained spectrum, a deeper understanding of the biological phenomena occurring during the progression/regression of multiple sclerosis can be elucidated, leading to the creation of more personalized and effective treatments.
