Highspeed 0.7 mm MAS probe for a 800 MHz solid state NMR spectrometer

Subject Area Basic Research in Biology and Medicine
Term Funded in 2018
Project identifier Deutsche Forschungsgemeinschaft (DFG) - Project number 418091302

Technical progress in solid-state NMR spectroscopy under magic angle spinning (MAS) has enabled significant improvement in sensitivity and resolution over the past few years. This methodology is essential for high-resolution studies on samples that are only available in sub-milligram amounts and cannot be obtained in deuterated form. Examples of such systems are proteins that cannot be expressed in eukaryotic systems or membrane proteins which cannot be refolded. Proton-detected experiments on such samples only reach the necessary resolution if MAS rates in the range above 100 kHz can be reached. These techniques were established in the past 3-4 years and only very recently have NMR probes become commercially available that enable such experiments. The possibility to conduct experiments on such equipment opens up completely new perspectives to research groups at BNMRZ addressing biological questions with solid-state MAS NMR.
