The impact of oxidative post-translational modifications on vascular function and angiocrine signaling (B01)

Project B01 will (i) characterize the angiocrine role of endothelial cell-derived S-nitrosated and S-sulfhydrated proteins on adipose tissue function during dietary-induced stress and aging, and (ii) analyze the mechanisms underlying any changes observed by determining alterations in gene expression, metabolic adaptation and function. Cellular and in vivo CSE and eNOS loss- and gain of function approaches will be pursued to study the impact of altered S-nitrosation and S-sulfhydration. The impact of S-nitrosation and S-sulfhydration on the secretion and shedding of multivesicular bodies, as well as on vesicle loading with oxPTM-modified cargo and their impact on adipocytes will be analyzed.
