In this project, we propose to investigate the repair of endogenous and induced DNA double-strand breaks during cell cycle progression using quantitative time-resolved analysis of individual living cells. To this end, we will combine live-cell imaging of fluorescent reporter cell lines with computation-al image processing and statistical analysis. Pharmacological perturbations of repair and signalling pathways will thus allow us to gain mechanistic insight into the consequences of repair pathways choices specifically during cell cycle transitions. We will use the resulting quantitative data to estab-lish mathematical models of the interplay between double-strand break repair and cell cycle progres-sion.
