Zusammenfassung der Projektergebnisse

After two years, our project results clearly indicate that enhancing Lmo4 levels in CD8+ T cells is a potent strategy for generating polyfunctional stem cell-like TCM cells. Moreover, these findings underline the therapeutic efficacy of methods targeted to enhance Lmo4 activity in CD8+ T cells. Unfortunately, we couldn’t demask any differences in stemness, memory formation and antitumor immunity by knock-out of Lmo4. The reason could be the physiologically very low expression of Lmo4 in CD8+ T cells, but also the possibility that other Lmo isoforms are compensating the potential changes in Lmo4∆/∆ T cells. In the same line, it was difficult to unveil a clear pathway which can explain the beneficial immune changes occurring after enforcing Lmo4 expression only by sequencing methods involving knock-out of Lmo4.
