The project focuses on the role of hemodynamic forces on and biomechanical properties of the vessel wall in the development of aortic disease with special emphasis on vascular inflammation modulated by purinergic signaling. Since biomechanical properties of the aorta depend on the respective aortic segment, we will comprehensively characterize the mouse aorta before and after induction of aortic stenosis or aneurysm with echocardiography and flow sensitive as well as molecular MRI. The developed protocols will be applied to specific mutant models for investigating the role of purinergic signaling in both disease models. In a translational step, we will verify these findings in patients with aortic stenosis and aneurysm.
