In this project we aim to identify a functional role for the lncRNA linc-PINT (p53-induced transcript) in vascular aging, remodeling and the development of abdominal aortic aneurysms. We identified linc-PINT as a potential key non-coding RNA regulating aneurysm formation and propose experiments using an organ-on-a-chip based system (aorta-on-a-chip) with patient-derived endothelial and smooth muscle cells, linc-Pint knockout mice and mechanistic molecular experiments. A potential therapeutic benefit for aneurysms will finally be tested in LDLR-deficient Yucatan mini-pigs. Overall, this project will show the role of linc-PINT in vascular disease.
