Pharmacological fMRI will be used in combination with a fully balanced placebo design to investigate psychological and neural mechanisms underlying the antidepressant effects of positive treatment expectation in patients with major depression, and how these effects combine or interact with acute antidepressant effects of the NMDA receptor antagonist esketamine. Prefrontal, limbic, and reward circuitry will be of special interest.
