At Saarland University several groups from the department of Organic Chemistry and Pharmacy (user community NMR) are involved in the isolation and total synthesis of complex natural products. Structure elucidation is generally accomplished via a combination of high resolution mass spectrometry and NMR spectroscopy. The last technique is essential not only for structure elucidation but also for the determination of the configuration of stereogenic centers, widespread found in natural products. In addition, binding studies of natural products or potential drugs towards their biological targets will be investigated (STD-NMR). With modern inverse cryoprobe technique the sensitivity of the spectrometer can be increased significantly, what is important if only small sample probes are available. The requested inverse probe head is especially sensitive for 1H nuclei, what is essential for structure and configuration determination. Such a probe head is not available to the user community so far. For reasons of economy we apply for a nitrogen- and not a helium-cooled probe head.The requested spectrometer is a substitution for a 25 year old 500 MHz spectrometer, which has to be replaced in due course. The building where it is located will be teared down this summer, and because of its unshielded magnet there is no possibility the run it somewhere else. In addition, software is completely obsolete and spare parts are not available any more.
