Triple-Quadrupol Mass spectrometer

Subject Area Medicine
Term Funded in 2020
Project identifier Deutsche Forschungsgemeinschaft (DFG) - Project number 453027627

The qualitative and quantitative analysis of chemical compounds is of central importance in drug research. The research-oriented Faculty of Pharmacy at the University of Bonn is broadly interconnected via the PharmaCenter Bonn, a Research Center for Innovative Drugs and Pharmacotherapy, and plays a central role in the area of life and health sciences. Hereby, we request a triple-quadrupol mass spectrometer for the platform „Chemical Analytics“ of the Faculty of Pharmacy for the sensitive quantitative analysis of small molecules. This MS instrument is urgently required for several, mostly third party-funded joint projects (e.g. GRK1873, FOR2372, FOR2625, FOR2685, SFB1328, TR-SFB259, TR-SFB261), since the currently available instruments are insufficient. A broad spectrum of analytical problems is to be handled, ranging from the quantitative determination of drug concentrations and metabolites in organoids and animal tissues, and the analysis of organic compounds in fossils, to innovative proteomics approaches. The platform for Chemical Analytics, which is associated with the Department of Pharmaceutical & Medicinal Chemistry, offers analytical service for the entire Faculty of Pharmacy, the PharmaCenter Bonn and beyond. The required mass spectrometer for quantitative analysis is complementary to the MS instruments that are available at the neighbouring, closely collaborating Chemical Institutes, which are focused on qualitative analysis and imaging. The new mass spectrometer will be utilized interdisciplinarily, between faculties and in the frame of (inter)national projects. Directly involved in the present application are besides the Department of Pharm. & Med. Chemistry (Müller, Gütschow), the Department of Pharmaceutical and Cell-Biological Chemistry (Hansen, Bendas), the Institute of Pharmaceutical Biology (König, Crüsemann), the Institute of Organic Chemistry (Menche, Engeser), and the Medical Faculty with the Institute of Molecular Biology and Biochemistry (Gieselmann, Winter) and the Institute of Pharmacology & Toxicology (Pfeifer). The envisaged user group, comprising high-ranking, experienced scientists as well as young talented scientists (Crüsemann, Engeser), will ensure complete occupancy of the instrument for work on sophisticated projects. Since students of Pharmacy are trained to become experts for Pharmaceutical Analytics including Instrumental Analytics, the MS instrument is also planned to be used for teaching (undergraduate research projects, and subsequent Master theses). Important criteria for the selection of the optimal system include (i) high sensitivity and linearity, (ii) flexibility, (iii) masses up to 2000 D, and (iv) determination of isomers.
