Gatsogiannis will use cryo-electron microscopy for a structure-function analysis of various key protein complexes in cell state transitions. He will (1) determine how substrate-engaged p97 cooperates with accessory factors involved in DNA repair and checkpoint control. He will (2) dissect a novel p97-HTRA1 complex that couples substrate unfolding and proteolysis. And (3), he will obtain structural insights into the molecular machinery that mediates and regulates mitotic spindle attachment to the chromosomal kinetochores to unravel how these factors co-operate to trigger metaphase to anaphase transition during cell division.
