We aim to harness γδ T cells for the treatment of BRAF-mutated colorectal cancer (CRC). In Aim 1, we will investigate how BRAF oncogenic signalling regulates the expression of γδ T-cell ligands and identify targetable molecules using pharmacological BRAF inhibition. In Aim 2, we will explore how BRAF signalling influences the tumour microenvironment and identify ways to overcome physical barriers limiting γδ T-cell infiltration. In Aim 3, we will engineer pro-infiltrating γδ T cells with enhanced anti-tumour activity. Our long-term goal is to develop combination therapies that improve γδ T-cell-mediated tumour control in CRC.
