Development of novel strategies to enhance adoptive T cell therapy for intracranial pediatric solid tumors by overcoming current obstacles and to improve patient outcomes: engineering of bispecific CARTs targeting B7H3+GD2 and B7H3+ROR2 to address antigen loss and tumor heterogeneity, design of FOXO1 co-expressing bispecific CARTs to enhance CART stemness, improving persistence and therapeutic efficacy, and to incorporate self-deactivating interleukin 12 (sdIL12) into bispecific CARTs to activate the innate immune system and enhance the anti-tumor response while minimizing toxicity.
