Exploitation of negative regulators of TCR signaling for therapeutic lymphocyte engineering (C01)

FYN was identified as a critical negative regulator of T-cell function and a putative regulator of PD-1, underscoring the therapeutic potential of modulating FYN activity in T cells. The spectrum of FYN’s role within TCR and CAR signaling, PD-1 checkpoint regulation, and possibly other ITIM/ITSM-containing receptors will be explored in detail to identify how FYN differentially governs these mechanisms and to explore the potential for selectively targeting FYN to enhance antitumor immunity.
