Enhancement of stemness, persistence, and efficacy of (CAR or TCR) T cell products through innovative metabolic-epigenetic engineering strategies. This will be anticipated and tested by modulating ACLY-mediated protein acetylation, optimizing amino acid utilization capacity, as well as targeting death receptor and ROS signaling pathways. In addition, it will be tested whether hypothermic conditions can be used to optimize CAR T cell products regarding cellular fitness, subset composition and functionality, and whether patient specific metabolic factors such as obesity and diabetes mellitus can affect the function and toxicity of CAR-T cell therapy.
