Subproject B01 covers the entire range from the detection of peri-implant infections to the autoregulatory reaction and thus has model character for project area B. Chemical and cellular sensor-actuator systems are being developed that release anti-biofilm agents via synthetic regulatory cassettes in response to infection-specific signals. To increase specificity, sensitivity, and therapeutic efficacy, cellular systems will be refined utilizing innovative signal amplification strategies and coupled with tailored chemical systems. Personalized 3D tissue-implant-biofilm models will be developed to enable in-depth characterization and validation of the systems, as well as to assess patient-specific factors influencing disease detection and treatment efficacy.
