We the rod photoreceptor active zone as an excellent model to 1) assess the molecular topology of Ca2+ channels and synaptic vesicle release sites, and 2) to study active zone function with electrophysiology. 1. In the third funding period, we will quantify numbers and relative topography of calcium channels, active zone proteins and synaptic vesicle proteins by 3D-MINFLUX, and we will test our hypothesis that calcium channels exert a nanodomain control on synaptic vesicle release.
