Lipid Dip-Pen Nanolithography for Model Bio-Membrane Systems

This aim of our collaboration supported by the NSF/DFG International Collaboration in Chemistry grant “Dip-Pen Nanolithography of Model Bio-Membranes and Membrane Stacks” was to demonstrate that Dip-Pen Nanolithography (DPN) is a unique tool for the fabrication of phospholipid membranes that are capable of mimicking cellular functions in vitro and to explore resulting possibilities for biological experiments. During the course of our work we found means for the controlled deposition of phospholipids with functional admixings by DPN. In the so called Lipid-DPN (L-DPN) DOPC was used as a carrier ink and by adjusting the environmental humidity and the substrate chemistry membranes of thicknesses from a single bilayer to hundreds of nanometers could be written. We showed that these assemblies are stable in cell culture and can be used to immobilize functional proteins. Based on these results the method of L-DPN was applied to the problem of immune activation and the response of mast cells to arrays of DNP (a potent allergen) produced by DPN were studied. An unexpected result of our research was the use of lipid gratings as biosensors, which was demonstrated as a label free detector system for streptavidin and will be enhanced in future applications. Additionally, the basic method of L-DPN was advanced further by establishing protocols for direct under water writing of lipids and demonstrating an easy way of monitoring the amount/mass deposited by L-DPN in particular lithographic features on the substrate.
