In the 2nd funding period, we have established the concept of a functional antagonism of OCaRs and NAADP-activated TPCs for the regulation of Ca2+ release from acidic Ca2+ stores. In the 3rd funding period, we plan to (i) investigate the role of the NAADP binding proteins HN1L/JPT2 and LSM12 as well as the relevance of OCaR1/OCaR2 within the NAADPsignaling pathway with global and local Ca2+ sensors, we aim (ii) to elucidate the role of TPCs/OCARs in the release of cytokines, (iii) to investigate the relevance of NAADP signaling pathways in cutaneous anaphylaxis and DSS colitis in in vivo models, and (iv) the reciprocal regulation of OCaRs after activation by mechanical stimuli.
