During the second funding period, we investigated how different synaptic states relate to plasticity in cortical neurons, focusing on silent synapses. In the third funding period, we propose to delineate the plasticity mechanisms that are gated by synapse idling. We aim to identify how new transmission sites are generated and unsilenced. We will focus on several mechanisms, including the role of astrocyte activation by Gq signaling, which is sufficient to trigger new silent synapses.
