Recombinant Negative Strand RNA viruses (NSV) are emerging as promising candidates for oncolytic virotherapy and other live virus applications. A disadvantage potentially jeopardizing applications of NSV is the lack of control options for RNA replication in critical tissue. We have started to evaluate the suitability of IRES elements to control rabies virus RNA replication. Here we suggest experiments towards the development of conditional (on/off) systems for RNA virus vectors based on IRESs. In addition, re-targeting and live imaging is being improved to study virus transport and delivery.
