The proposed project plans the comprehensive analysis of the structure-function relationships of LG modeules of laminin a1 to a5 chains and of the proteoglycan perlecan. These LG modules will be obtained by recombinant production in mammalian cells and used for the mapping of binding sites important for cellmatrix interactions and the supramolecular organization of basement membranes. The ligands examined will be integrin and a-dystroglycan receptors, bacterial receptors, sulfatides, heparin/heparan sulfate and some matrix proteins. Site-directed mutagenesis and X-ray structural analyses will be used for a precise mapping of binding epitopes on various LG modules.
