The research underlying the projects described covers a wide range of quantitative and qualitative research questions related to the analysis of modified and unmodified peptides. A further focus is the time-resolved detection of small molecule metabolites. Both classes of analytes are predestined for targeted mass spectrometric studies and for their unequivocal detection, quantification and publication-relevant characterization, liquid chromatography coupled to tandem mass spectrometry based on a triple quadrupole configuration is currently the gold standard due to ist excellent sensitivity. The technology allows the highly specific, parallel quantification of target molecules in complex reaction mixtures over a dynamic range of up to more than six orders of magnitude from the femto- to the nanogram range. It is in particular unrivalled for kinetic measurements such as time resolved expression analyses of complex protein samples or post-translational modifications. The optional coupling of the mass spectrometer unit to an ultra-high-performance liquid chromatography system or to a nano-liquid chromatography system allows the use of the mass spectrometer for project-related routine measurements as well as for more specific analyses of samples from limited biological source material. A triple quadrupole mass spectrometer coupled with liquid chromatography is currently not available at the Department of Chemistry and Chemical Biology at TU Dortmund University and, due to ist broad applicability in various areas of biochemistry, chemical biology and organic chemistry, represents an imperative addition to the analytical instrumentation available at the site
