The epidermis is mainly composed of keratinocytes, which provide a physical barrier and also play a key role in skin immunity. We have discovered a new inflammasome induced by NLRP10 in highly differentiated keratinocytes in the stratum granulosum of the skin. NLRP10 can form an inflammasome upon the demise of mitochondria and trigger a new form of inflammasome-dependent anti-inflammatory cell death. Our future research will focus on identifying factors involved in NLRP10 inflammasome activation, defining the mechanisms of cell death triggered by NLRP10 activation, and assessing the in vivo relevance of NLRP10 in atopic dermatitis and skin homeostasis.
