H/ACA-complexes catalyze the pseudouridylation of numerous cellular RNAs. The underlying mechanisms as well as the sequence-specific recognition of substrate RNAs are however insufficiently understood. This also holds true for the effect of disordered protein domains, their posttranslational modification, and their role in RNA binding and phase separation. Using a combination of biochemical and analytical tools together with single molecule FRET spectroscopy, we will characterize the underlying mechanisms and structural dynamics. In this project, we will investigate the selection and catalytic modification of substrate RNAs as well as the role of individual protein domains, generating a new interface between RNA modifications and posttranslational protein modifications.
