Entry into cell cycle in late G1 is a central regulatory step of the eukaryotic cell cycle. In yeast, the timing of S-phase entry is largely determined by transcriptional activation of G1 cyclins und DNA synthesis genes as cells reach a critical size. Their expression is regulated by the cyclin dependent kinase Cdc28p and depends on two transcription factor complexes called SBF (Swi4/Swi6) and MBF (Mbp1/Swi6). In the proposed project we study the function and regulation of these transcription factors. We have genetically and biochemically identified novel proteins (Ctr9p and Paf1p) required for gene expression in late G1. Their phyiological role is being analysed. The aims of the project are to elucidate how transcriptional activation in late G1 is coordinated with cell size and cellular growth.
