In morphological terms there are clear differences between the early secretory pathways of mammalian, yeast and plant cells. The research proposed here is directed towards uncovering the molecular basis for the differences in plant cells, and is channelled into three interdependent sub-projects: - an elucidation of the mechanism of ER-export with particular emphasis on identifying, isolating and characterzing the (putative COPII-) vescles involved - an examination of the parameters influencing retograde Golgi>ER transport, and attempts at reconstituting this process in vitro using the assembly of Golgi-derived IgG heavy chain-HDEL with ER-based IgG light chains as a measure for succesful vesicle fusion - an analysis of the events leading to reconstitution of the Golgi apparatus in cell recovering from the COPI-vesculation inhibitor brefeldin A (BFA).
