Neighboring cells can differ dramatically with respect to metabolism. Furthermore, metabolism is highly compartmentalized. To understand metabolic interactions of cells and compartments, it is essential to be able to determine the actual concentrations of metabolites in vivo and to follow their changes in response to stimuli in real time with subcellular resolution. We developed a nanosensor that can be used to measure maltose in vivo and it is based on a periplasmic binding protein coupled to two different fluorescent proteins. The aim of this project is to optimize the sensor, to use this sensor for in vivo measurements in plants and to develop novel nanosensors for glucose and ribose on the basis of structurally related periplasmic binding protein moieties.
