Functional and structural characterization of bacterial blue-light sensitive photoreceptors
Zusammenfassung der Projektergebnisse
Im TP A1 wurden bakterielle Phototropin-ähnliche Photorezeptoren untersucht. Beginnend mit dem ersten prokaryotischen Vertreter dieser Substanzklasse, YtvA aus B. subtilis, wurden spektroskopische, kinetische und funktionelle Experimente zum Verständnis der molekularen Reaktivität und zur Erzeugung und Weiterleitung des biologischen Signals durchgeführt. Im weiteren Verlauf wurden, aufbauend auf Datenbank-Suchen, Gene für neuartige bakterielle LOV-Domänen Proteine identifiziert, kloniert und deren rekombinante Genprodukte funktionell charakterisiert. Besonderes Gewicht wurde hierbei auf die Demonstration lichtinduzierter Regulation ankondensierter Enzymaktivitäten gelegt. Abschliessend wurden die Arbeiten auf Metagenom-Analysen und funktionelle Charakterisierung möglicher LOV-Domänen aus diesen Daten ausgedehnt.
