Translation of certain defective mRNAs can cause abnormally long pauses in ribosome elongation, resulting in truncated proteins that are likely to be dysfunctional. The ribosome associated quality control (RQC) pathway addresses these ribosome-stalled proteins for degradation, limiting their toxic effects. In the project we will systematically characterize defective transcripts arising upon proteotoxic stress at the ER, as well as the impact of RQC on proteome fidelity. By integrating transcriptomic and proteomic data, we envision obtaining a mechanistic understanding of the physiological causes and consequences of RQC.
