Ring finger protein 213 (RNF213) contains an active E3 ligase domain and plays a role in modulating cell-intrinsic inflammation and vascularization. RNF213 is expressed in different splicing isoforms whose abundance and localization vary across the lung cancer microenvironment, and these variations are associated with overall patient survival. In the proposed project, our aim is to define the ubiquitylation substrates and molecular and cellular functions of RNF213 and its variants. Focusing on the role of RNF213 variants in lung cancer, we aim to identify novel RNF213-dependent cancer vulnerabilities that may be targeted to modulate lung cancer progression.
