In B07, Elmar Wolf and Gabriele Büchel will target the Aurora-A complexes and their oncogenic function in AML by PROTACs. First, they will investigate which non-substrate proteins directly bind Aurora-A in AML, the structural nature of these interactions, and the effects of PROTAC-induced depletion of Aurora-A on these protein complexes. Using auxin models (ubiquitin-based DEGRON systems), they will analyze the phenotype of Aurora-A interaction partner degradation. Second, they will test the therapeutic potential of Aurora-A degradation in AML mouse models. In addition, they aim to identify biomarkers for the use of Aurora-A PROTACs in AML patients.
