The spatial arrangement of chromatin in the nucleus is crucial for correct gene expression. The 3D folding of chromatin is influenced by the cohesin complex but also the underlying 1D nucleosome positioning, as we could recently show in S. cerevisiae. Nucleosome positioning can regulate gene expression and is determined mainly by ATP-dependent chromatin remodeling enzymes (remodelers). Here we want to decipher how cohesin and remodelers interplay to regulate 3D genome folding and gene expression by using an unique genome-wide reconstitution approach.
