Access to quality-controlled, well-documented bacterial strains will be essential to achieve the main objective of SPP 2474 to unlock the functional potential within the human gut microbiome. Most of the SPP projects specifically require novel isolates of selected target species, defined synthetic consortia, and genome-wide mutant libraries. Additional needs are genome sequencing, in-depth strain typing and characterization, long-term deposit of defined strain collections, and compliance with Access and Benefit Sharing regulations. The Z01-project combines the complementary expertise of two groups in these areas and thereby offers a centralized, specific catalogue of services to support all SPP projects with cultivation-based analyses of microbiomes. Anaerobic cultivation services that require a broad range of methods and elaborate hardware will be provided to SPP members, enabling the isolation and subsequent characterisation of multiple strains of model bacterial species and also of novel taxa. Isolates will undergo genome-based taxonomic and functional analysis, and tailored phenotyping according to the specific needs of individual projects. A novel strain deposition process will be implemented to secure all relevant strains generated within the SPP for future studies and to accommodate bulk submissions of large strain collections, including transposon libraries. Additional services include project-specific design of synthetic communities of cultured bacteria (syncoms), the expansion of phenotypic databases with detailed information on all strains offered, and the creation of web interfaces that enable specific search and data integration functions for SPP projects. Through these comprehensive approaches, Z01 will catalyse the research within individual projects, generate sustainable added value for the whole community, and thereby increase the visibility of SPP2474 worldwide.
