Intestinal L cells release the hormone glucagon-like peptide-1 (GLP-1) in response to ingestion of monounsaturated fat, such as found in olive oil. GLP-1 then increases insulin secretion and reduces blood sugar levels. Additionally, GLP-1 decreases both feelings of hunger and body weight. Previously, we have shown the involvement of an enzyme Protein kinase Czeta (PKCzeta) in GLP-1 secretion in isolated cells, but the role of this enzyme in GLP-1 secretion in living organisms remains unknown. Therefore, by reducing expression of PKCzeta in the intestinal L cells in rats, we will determine the relationship between monounsaturated fat acids, PKCzeta and GLP-1 secretion. Two GLP-1-related drugs have already been approved in the USA and Europe for the treatment of patients with type 2 diabetes. The stimulation of L cells to increase GLP-1 secretion may be an additional therapeutic approach.
