Our project will tackle three key questions: (1) How does the anatomy of the tripartite synapse look like at the nanoscale in living brain slices, i.e., free of fixation artifacts? (2) How does this nano-anatomy shape the transient bio-availability of signaling substances in the extracellular space in and around tripartite synapses? (3) How does synaptic activity affect or remodel the nano-anatomy of the tripartite synapse, and in turn impact the bio-availability of signaling substances, and thereby synaptic function?
