The energy metabolism plays a crucial role in a wide variety of vital cellular processes, and studies of metabolic pathways are used in numerous areas of research. In practically all organs, the physiological partial pressure of oxygen (pO2) is below the atmospheric pO2 (at sea level) of 159 mmHg (20.9 % O2) and below the arterial pO2 of 100 mmHg. In most tissues, the pO2 is physiologically and under pathophysiological conditions much lower (in tissue surrounding average capillaries the pO2 ranges physiologically from 75 mmHg at the arterial end all the way down to 7.5 mmHg at the venous end). Nonetheless, most studies perform their investigations at atmospheric pO2 levels, which does not reflect the local tissue conditions. It is absolutely necessary to perform metabolic analyses of different cell types at oxygen partial pressures that correspond to the local environment in tissues under (patho-)physiological conditions. In order to improve and strengthen the possibilities and relevance of research into the energy metabolism at the Greifswald research site, it is therefore essential to acquire an extracellular flux analyser combined with a specifically designed hypoxia chamber with control of atmospheric oxygen levels.
