Differential roles of resident microglia and myeloid cells in neuroinflammatory conditions (A07)

The project will characterize the cellular and molecular mechanisms of tissue scarring in the CNS. This group will use novel transgenic mouse models and intravital two-photon microscopy to visualize the interactions of pericytes and fibrogenic cells with resident microglia and invading myeloid cells in EAE and cerebral ischemia (MCAO). In order to identify the molecular signals linking fibrotic and inflammatory responses, they will perform RNA sequencing in stromal cells, microglia and peripherally derived innate immune cells. Finally, they will determine the impact of inflammation on tissue scarring in the CNS by specific cell ablation of microglia and peripheral myeloid cells, as well as by deletion of key mediator genes such as TGF-ß1.
