Ca2+-permeable channels as targets and regulators of Ca2+-dependent phosphorylation

Ca2+-permeable ion channels are important components of Ca2+-dependent signalling cascades. They form pathways for Ca2+ entry into the cytosol and they represent targets for Ca2+-dependent regulation. This project investigates the regulation of cation channels in the plasma and vacuolar membrane by Ca2+ and Ca2+-dependent phosphorylation. Cyclic nucleotide gated channels (CNGC) in the plasma membrane and the two-pore channel TPC1 in the vacuolar membrane serve as model systems. Our collaborations within the Research Unit will characterize the mechanism and role of TPC1 regulation by the Ca2+-regulated kinase CIPK1, identify other possible interacting kinases of CNGCs, study the mechanisms of interaction, and examine the impact of the interactions on cellular functions. Using molecular, electrophysiological and cell biological approaches, the physiological role of the two channels and their regulation by Ca2+ and Ca2+-dependent kinases will be addressed.
