Interference of the viral effector proteins pp71 and IE1 with ND10-mediated intrinsic immunity against human cytomegalovirus infections (B03)

We have demonstrated that a specific organelle of the cell nucleus, termed PML nuclear body (PML-NB), is modified by viral effector proteins such as the pp71- or IE1-protein of human cytomegalovirus. This correlates with antagonization of a direct repressive function of PML-NB components that act as cellular restriction factors. Recent studies reveal an emerging role of PML-NBs as co-regulatory structures of further innate responses (e.g. IFN pathway). This proposal investigates how IE1 and pp71 are able to evade intrinsic and innate immune responses particularly governed by TRIM factors.
