Sodium/proton exchangers are key players in the essential control of intracellular pH and cell volume. The precise modulation of their transport activity is the prerequisite for their distinct functions in a broad spectrum of physiological processes. Their dysfunction is implicated in several diseases including epilepsy, cardiac hypertrophy and cancer. We aim for the elucidation of mechanism and structural basis of sodium/proton exchange activity modulated by the interaction with regulatory proteins and covalent modification with emphasis on the link to Ca2+ signaling to contribute to a detailed understanding of the specific functions of sodium/proton exchangers in health and disease.
